1p13.2 deletion displays clinical features overlapping Noonan syndrome, likely related to NRAS gene haploinsufficiency Genet. Mol. Biol.
Linhares,Natália Duarte; Freire,Maíra Cristina Menezes; Cardenas,Raony Guimarães Corrêa do Carmo Lisboa; Pena,Heloisa Barbosa; Lachlan,Katherine; Dallapiccola,Bruno; Bacino,Carlos; Delobel,Bruno; James,Paul; Thuresson,Ann-Charlotte; Annerén,Göran; Pena,Sérgio D. J..
Abstract Deletion-induced hemizygosity may unmask deleterious autosomal recessive variants and be a cause of the phenotypic variability observed in microdeletion syndromes. We performed complete exome sequencing (WES) analysis to examine this possibility in a patient with 1p13.2 microdeletion. Since the patient displayed clinical features suggestive of Noonan Syndrome (NS), we also used WES to rule out the presence of pathogenic variants in any of the genes associated with the different types of NS. We concluded that the clinical findings could be attributed solely to the 1p13.2 haploinsufficiency. Retrospective analysis of other nine reported patients with 1p13.2 microdeletions showed that six of them also presented some characteristics of NS. In all...

5S rDNA characterization in twelve Sciaenidae fish species (Teleostei, Perciformes): depicting gene diversity and molecular markers Genet. Mol. Biol.
Alves-Costa,Fernanda A.; Martins,Cesar; Matos,Fernanda Del Campos de; Foresti,Fausto; Oliveira,Claudio; Wasko,Adriane P..
In order to extend the genetic data on the Sciaenidae fish family, the present study had the purpose to characterize PCR-generated 5S rDNA repeats of twelve species of this group through PAGE (Polyacrylamide Gel Electrophoresis) analysis. The results showed the occurrence of at least two different 5S rDNA size classes in all the species. Moreover, 5S rDNA repeats of one of the studied species - Isopisthus parvipinnis - were cloned and subjected to nucleotide sequencing and Southern blot membrane hybridization analyses, which permitted to confirm the existence of two major 5S rDNA classes. Phylogenetic analysis based on the nucleotide sequences of different 5S rDNA repeats of I. parvipinnis lead to their separation into two major clusters. These results may...

-866G/A and Ins/Del polymorphisms in the UCP2 gene and diabetic kidney disease: case-control study and meta-analysis Genet. Mol. Biol.
Dieter,Cristine; Assmann,Taís Silveira; Lemos,Natália Emerim; Massignam,Eloísa Toscan; Souza,Bianca Marmontel de; Bauer,Andrea Carla; Crispim,Daisy.
Abstract Uncoupling protein 2 (UCP2) decreases reactive oxygen species (ROS). ROS overproduction is a key contributor to the pathogenesis of diabetic kidney disease (DKD). Thus, UCP2 polymorphisms are candidate risk factors for DKD; however, their associations with this complication are still inconclusive. Here, we describe a case-control study and a meta-analysis conducted to investigate the association between UCP2 -866G/A and Ins/Del polymorphisms and DKD. The case-control study comprised 385 patients with type 1 diabetes mellitus (T1DM): 223 patients without DKD and 162 with DKD. UCP2 -866G/A (rs659366) and Ins/Del polymorphisms were genotyped by real-time PCR and conventional PCR, respectively. For the meta-analysis, a literature search was conducted...

A antracnose do sorgo Trop. Plant Pathol.

A biogeographical population genetics perspective of the colonization of cats in Latin America and temporal genetic changes in Brazilian cat populations Genet. Mol. Biol.
Ruiz-García,Manuel; Alvarez,Diana.
We used nine morphological genes to analyze cat populations from Mexico, the Dominican Republic, the Colombian, Brazilian and Peruvian Amazon, Bolivia and Brazil. Most populations were in Hardy-Weinberg equilibrium at the O locus. The highest allele frequencies so far detected at world level for alleles I (inhibitor) and L (long hair) were found at La Paz (Bolivia). The analyses revealed at least five cat gene pools in Latin America: These findings suggest that the current genetic distribution of cats in Latin America correlates with the colonization of the Americas during the XIV to XVIII centuries. Additionally, the cat populations of São Paulo, Rio de Janeiro and Manaus were sampled, to compare their 1996-2003 genetic profiles with those obtained in...

A canonical correlation analysis of the association between carcass and ham traits in pigs used to produce dry-cured ham Genet. Mol. Biol.
Ventura,Henrique T.; Lopes,Paulo S.; Peloso,José V.; Guimarães,Simone E.F.; Carneiro,Antonio Policarpo S.; Carneiro,Paulo L.S..
The association between carcass and ham traits in a pig population used to produce dry-cured ham was studied using canonical correlation analysis. The carcass traits examined were hot carcass weight (HCW), backfat thickness (BT) and loin depth (LD), and the ham traits studied were gross ham weight (GHW), trimmed ham weight (THW), ham inner layer fat thickness (HIFT), ham outer layer fat thickness (HOFT), pH (pH) and the Göfo value. Carcass and ham traits are not independent. The canonical correlations (r) between the carcass and ham traits at 130 kg were 0.77, 0.24 and 0.20 for the first, second and third canonical pair, respectively, and were all significant (p < 0.01) by the Wilks test. A case of albinism in the Central American spider monkey, Ateles geoffroyi, in Honduras Mastozool. neotrop.
Espinal,Mario; Mora,José Manuel; Ruedas,Luis A; López,Lucía Isabel; Marineros,Leonel.

A case of mosaicism in the germinal line of Odontophrynus americanus (Dùmeril et Bibron. 1841) JBAG
Salas,Nancy; Martino,Adolfo.
The cytogenetic studies have special importance in the zoological research because by means of karyotype analysis they contribute to the identifi cation of different entities, facilitate the evaluation of the proximity or relationship degree among certain species and fi nally they contribute with fundamental data to the phylogenetic assessment (Schmid et al., 1985; Zampieri et al., 2004). The occurrence of polyploidy in different anuran families that appear as cryptic species highlights the importance of the cytogenetic study at the population level (Valetti et al., 2009).

A case of prenatal diagnosis of fetal hydantoin syndrome by ultrasound Genet. Mol. Biol.
Gollop,Thomaz Rafael; Salzo,Ivan.
Fetal hydantoin syndrome (FHS) is a set of disruptions occasionally present in fetuses exposed in utero to phenytoin or other anticonvulsants. Administration of phenytoin in early pregnancy may impair proper psychomotor performance expected for children's development. Several combined phenotypic markers delineate the syndrome, but the presence of single clinical signs is more common. There is controversy about the etiology of FHS. Associated disruptions may be related to a deficiency in a detoxifying enzyme (epoxide hydrolase), vascular problems, and/or factors not yet known. Genetic causes are believed to influence susceptibility to the drug. This text reports an unusual pattern of malformations detected in an ultrasound scan (gastroschisis, sacral...

A comparative study of eleven protein systems in tamarins, genus Saguinus (Platyrrhini, Callitrichinae) Braz. J. Genet.
The genetic variability of six tamarin taxa, genus Saguinus, was analyzed comparatively using protein data from eleven systems coded by 15 loci. S. fuscicollis weddelli and S. midas midas were the most polymorphic taxa, and S. bicolor the least. The results of the phylogenetic analyses (UPGMA and neighbor-joining) and the genetic distances between taxa were generally consistent with their geographic and probable phylogenetic relationships. Analyses of the S. bicolor and S. midas populations suggested that they represent no more than three subspecies of a single species, S. midas, with the bicolor forms belonging to a single subspecies, S. midas bicolor. If supported by additional studies, this would have important implications for the conservation of the...

Abstract Next-generation sequencing (NGS) platforms allow the analysis of hundreds of millions of molecules in a single sequencing run, revolutionizing many research areas. NGS-based microRNA studies enable expression quantification in unprecedented scale without the limitations of closed-platforms. Yet, whereas a massive amount of data produced by these platforms is available, comparisons of quantification/discovery capabilities between platforms are still lacking. Here we compare two NGS-platforms: SOLiD and PGM, by evaluating their microRNA identification/quantification capabilities using two breast-derived cell-lines. A high expression correlation (R2 > 0.9) was achieved, encompassing 97% of the miRNAs, and the few discrepancies in miRNA counts were...

A comparison of metrics for estimating phylogenetic signal under alternative evolutionary models Genet. Mol. Biol.
Diniz-Filho,José Alexandre F.; Santos,Thiago; Rangel,Thiago Fernando; Bini,Luis Mauricio.
Several metrics have been developed for estimating phylogenetic signal in comparative data. These may be important both in guiding future studies on correlated evolution and for inferring broad-scale evolutionary and ecological processes (e.g., phylogenetic niche conservatism). Notwithstanding, the validity of some of these metrics is under debate, especially after the development of more sophisticated model-based approaches that estimate departure from particular evolutionary models (i.e., Brownian motion). Here, two of these model-based metrics (Blomberg's K-statistics and Pagel's λ) are compared with three statistical approaches [Moran's I autocorrelation coefficient, coefficients of determination from the autoregressive method (ARM), and phylogenetic...

A comparison of the effects of physical and chemical mutagens in sesame (Sesamum indicum L.) Genet. Mol. Biol.
Begum,Tamina; Dasgupta,Tapash.
Three sesame genotypes (Rama, SI 1666 and IC 21706) were treated with physical (γ-rays: 200 Gy, 400 Gy or 600 Gy) or chemical (ethyl methane sulphonate, EMS: 0.5%, 1.0%, 1.5% or 2.0%) mutagens and their mutagenic effectiveness and efficiency were estimated in the M2 generation. The M3 generation was used to identify the most effective mutagen and dose for induction of mutations. The average effectiveness of EMS was much higher than γ-rays. The lowest dose of γ-rays (200 Gy) and the lowest concentration of EMS (0.5%) showed the highest mutagenic efficiency in all genotypes. Analysis of the M3 generation data based on parameters such as the variance ratio and the difference in residual variances derived from the model of Montalván and Ando indicated that...

A comparison of the hygienic response of Africanized and European (Apis mellifera carnica) honey bees to Varroa-infested brood in tropical Brazil Genet. Mol. Biol.
Aumeier,Pia; Rosenkranz,Peter; Gonçalves,Lionel Segui.
In order to examine the significance of hygienic behavior for the tolerance to varroosis of Africanized honey bees, they were compared with non-tolerant Carniolans in tropical Brazil. Capped worker brood cells were artificially infested with living Varroa mites, and inspected some days later. Uncapping, disappearance of the introduced mite and removal of the pupa were recorded in a total of manipulated 3,096 cells during three summer seasons. The hygienic response varied between Africanized and Carniolan colonies, but this difference was significant only in one year, during which Africanized honey bees removed a significantly greater proportion of Varroa mites than European honey bees. A high proportion of the mites disappeared from artificially infested...

A comprehensive transcriptome analysis of skeletal muscles in two Polish pig breeds differing in fat and meat quality traits Genet. Mol. Biol.
Piórkowska,Katarzyna; Żukowski,Kacper; Ropka-Molik,Katarzyna; Tyra,Mirosław; Gurgul,Artur.
Abstract Pork is the most popular meat in the world. Unfortunately, the selection pressure focused on high meat content led to a reduction in pork quality. The present study used RNA-seq technology to identify metabolic process genes related to pork quality traits and fat deposition. Differentially expressed genes (DEGs) were identified between pigs of Pulawska and Polish Landrace breeds for two the most important muscles (semimembranosus and longissimus dorsi). A total of 71 significant DEGs were reported: 15 for longissimus dorsi and 56 for semimembranosus muscles. The genes overexpressed in Pulawska pigs were involved in lipid metabolism (APOD, LXRA, LIPE, AP2B1, ENSSSCG00000028753 and OAS2) and proteolysis (CST6, CTSD, ISG15 and UCHL1). In Polish...

A critical evaluation of the effect of population size and phenotypic measurement on QTL detection and localization using a large F2 murine mapping population Genet. Mol. Biol.
Li,Xinmin; Quigg,Richard J; Zhou,Jian; Xu,Shizhong; Masinde,Godfred; Mohan,Subburaman; Baylink,David J..
Population size and phenotypic measurement are two key factors determining the detection power of quantitative trait loci (QTL) mapping. We evaluated how these two controllable factors quantitatively affect the detection of QTL and their localization using a large F2 murine mapping population and found that three main points emerged from this study. One finding was that the sensitivity of QTL detection significantly decreased as the population size decreased. The decrease in the percentage logarithm of the odd score (LOD score, which is a statistical measure of the likelihood of two loci being lied near each other on a chromosome) can be estimated using the formula 1 - n/N, where n is the smaller and N the larger population size. This empirical formula has...
